What paternity testing actually does
A paternity test compares the child's DNA with the DNA of one or more possible fathers. We all inherit half our DNA from our biological mother and half from our biological father, so a true father's DNA will match the child at every tested marker. If enough markers do not match, paternity is excluded.
Standard testing looks at about 15-25 short tandem repeat (STR) markers and gives a very confident answer: usually above 99.9% probability when a man is the father, and essentially 0% when he is not. After birth the test is simple, just a cheek swab from the baby and the man. The real challenge is doing it during pregnancy, because the baby's DNA is not easy to reach safely. The three ways to test paternity before birth
- Non-invasive prenatal paternity testing (NIPP)
During pregnancy, small fragments of the baby's DNA (called cell-free fetal DNA) circulate in your bloodstream, shed naturally by the placenta. This is the same biology used by NIPT, the non-invasive prenatal test for chromosomal conditions. CVS is an invasive procedure done between 10 and 13 weeks, in which a thin needle or catheter takes a tiny sample of placental tissue under ultrasound guidance. It carries roughly a 0.5-1% procedure-related miscarriage risk, so it is almost never done for paternity alone.

How non-invasive prenatal paternity testing (NIPP) works
If you are weighing NIPP through an international service, it helps to understand the mechanics.
The biology. The placenta constantly renews itself, releasing short fragments of fetal DNA into your blood. Early on, around 7-8 weeks, the baby's DNA makes up only about 5-10% of the cell-free DNA in your plasma (the "fetal fraction"), rising to 10-20% or more by mid-pregnancy. A higher fetal fraction gives a more reliable result, which is why testing too early can occasionally fail and need repeating.
What is collected. A venous blood sample from you (about 10-20 mL) and a DNA sample from the alleged father (usually a cheek swab). If the result might be used legally, both samples must be collected with a documented chain of custody.
In the lab. The cell-free DNA is sequenced, the baby's DNA is separated from yours based on inheritance patterns, and thousands of genetic markers (SNPs) are compared with the father's DNA to calculate a probability of paternity. Results, usually in 1-3 weeks, report a probability approaching 99.9% for a match or near 0% for an exclusion.
When results may be unclear. The most common reason is too little fetal DNA, fixed by retesting a little later. Less commonly, poor sample quality, certain twin pregnancies, a recent blood transfusion or organ transplant, or some maternal illnesses can interfere.
Accreditation matters. Trustworthy NIPP comes from AABB-accredited (or equivalent) laboratories with validated methods and proper quality control. Be wary of marketing-led services without rigorous accreditation.
Invasive testing: CVS and amniocentesis
Both CVS and amniocentesis can give paternity results equal in accuracy to a postnatal test, because they sample the baby's own cells directly. But they exist to diagnose genetic and chromosomal conditions, not to answer paternity questions, and they carry a small but real miscarriage risk.
CVS, between 10-13 weeks, takes 5-20 mg of placental villi. Amniocentesis, from 15 weeks, draws 15-30 mL of amniotic fluid. Both are done under ultrasound guidance by experienced fetal-medicine specialists. Both carry roughly a 0.5-1% procedure-related miscarriage risk (lower in high-volume centres), with rare risks of infection, bleeding, or fluid leak. These procedures are usually performed when there is a genuine clinical reason, such as a high-risk screening result or carrier status for an inherited condition like thalassemia or sickle cell. Choosing an invasive procedure solely for paternity means accepting a real pregnancy-loss risk for a question that postnatal testing answers safely, which is why it is essentially never done this way in routine Indian practice.
Postnatal DNA testing: the Indian standard
For most families, the simplest and best path is to wait until after birth.
The sample is just a painless cheek swab from the baby and the alleged father (cord blood collected at delivery also works). Swabs stay stable for weeks at room temperature, so there is no rush. A lab compares 15-25 STR markers and reports a probability of paternity, typically over 99.99% when a man is the father, and essentially 0% when he is not. Accredited testing is over 99.9% accurate; genuine errors are almost always sample mix-ups, not technical failures.
In India, postnatal paternity testing costs about INR 8,000-25,000, and is offered by NABL-accredited DNA laboratories and major hospital chains across metro cities. For legal use, the test must be done with verified identity and a documented chain of custody; a cheaper "peace-of-mind" test uses the same technology but is not admissible in court.
